As a best practice, enrollments should always be setup at the course level as a ruleset enrollment instead of on a group at the group level. On a course level enrollment, you are able to add multiple rulesets to the same enrollment. This is easier for the system to process than an enrollment that is applied to each individual group. The system will scan the properties page of a course level ruleset enrollment and apply the enrollment to all users that fall into the rules of that enrollment.
Enrollments should be applied to user accounts that meet the ruleset criteria within 3-5 minutes of the ruleset being created and saved. If you are noticing a delay with enrollments, if could be that your rules are built incorrectly OR that you have too many rulesets running throughout your site that need to be reviewed and cleaned up. The more rulesets running throughout your system, the longer this delay will be.
